uniQure’s Huntington’s Gene Therapy Wins FDA Acceptance and Priority Review, Setting Up a May 2027 Decision
uniQure N.V. (NASDAQ: QURE) cleared a major regulatory hurdle on Monday. The FDA accepted for review the company’s Biologics License Application (BLA) for ifezuntirgene inilparvovec (AMT-130), its investigational gene therapy for Huntington’s disease, which uniQure submitted under the Accelerated Approval pathway. The agency also granted Priority Review and set a target action date of May 1, 2027.
Key Points
- If approved, ifezuntirgene inilparvovec would be the world’s first disease-modifying treatment for Huntington’s disease.
- The FDA said it tentatively plans to discuss the application at an Advisory Committee meeting.
- The therapy was the first Huntington’s candidate to receive both Breakthrough Therapy and Regenerative Medicine Advanced Therapy (RMAT) designations, and it also holds Fast Track designation.
- The milestone strengthens uniQure’s standing in the PRISM Biotech Index.
What the FDA Accepted From uniQure (NASDAQ: QURE)
The application rests on long-term data. It draws on 36-month results from uniQure’s Phase I/II study, compared against a propensity score-matched external control drawn from the Enroll-HD natural history database. In total, the Phase I/II program has dosed 51 patients across two multi-center studies.
The therapy itself is a one-time treatment. It uses uniQure’s gene-silencing miQURE® platform to silence the huntingtin gene and a potentially highly toxic protein fragment. Surgeons deliver a single dose directly into the striatum using MRI-guided neurosurgery.
A Long Road to This Point
Today’s acceptance caps a volatile stretch for the stock. PRISM MarketView has tracked each turn, from uniQure’s shares plunging after the FDA rejected key gene therapy data, to the stock surging 36% after a key FDA leadership departure. Then, in June, uniQure jumped as the FDA cleared a faster path for its Huntington’s gene therapy.
CEO Matt Kapusta called the acceptance a major milestone for a Huntington’s community that has waited generations for a treatment that changes the course of the disease.
The Confirmatory Trial Is Already Planned
uniQure and the FDA have agreed on the design of a confirmatory study. The randomized trial will enroll about 200 patients, assigned two to one between the gene therapy and standard of care, with Total Functional Capacity measured at 36 months as the primary endpoint. Patient screening in the U.S. is set to begin by the end of 2026, and Kapusta expects the study to be well underway by the time of a potential approval.
Why It Matters for the PRISM Biotech Index
The unmet need is enormous. About 75,000 people live with Huntington’s disease across the U.S., EU and UK, and hundreds of thousands more are at risk of inheriting it. Despite that, no approved therapy currently delays the onset of the disease or slows its progression.
What to Watch Next for uniQure
The next catalysts are the Advisory Committee date, which uniQure says it will learn once the FDA finalizes timelines, and the start of confirmatory trial screening by year-end.
