Oncolytics Says Every Responder Survived Past One Year in Anal Cancer Study, Sets Sights on HPV-Driven Head and Neck Cancer
Oncolytics Biotech® Inc. (Nasdaq: ONCY) today released updated durability and survival data showing that every patient who responded to its pelareorep combination in previously treated anal cancer lived beyond 12 months. The company now plans to use those results as the foundation for a broader immunotherapy platform targeting cancers driven by human papillomavirus (HPV).
Key Points
- Pelareorep plus atezolizumab (Tecentriq®) delivered a median duration of response of roughly 15 months in second-line or later squamous cell anal cancer (SCAC), compared with 9.5 months historically reported for FDA-approved retifanlimab monotherapy.
- All responding patients survived longer than one year, and several have now survived beyond two years; median overall survival has not yet been reached.
- Oncolytics will explore additional studies with a potential checkpoint inhibitor partner to support registration in SCAC and may expand into HPV-associated head and neck cancer.
Durable Responses in a Hard-to-Treat Population
The new figures come from Cohort 4 of the Phase 1/2 GOBLET study, which tests pelareorep with Roche’s checkpoint inhibitor atezolizumab in patients whose anal cancer progressed after earlier treatment. Patients in this setting currently have few options, and more than 90% of SCAC cases are linked to HPV.
According to the company’s internal calculations, responders on the combination stayed in response for about 15 months on average, well ahead of the 9.5-month benchmark set by the only approved immunotherapy in a comparable population. Moreover, because median overall survival has not yet been reached, the survival picture may continue to strengthen as patients are followed longer.
The update builds on momentum in the program. In July, Oncolytics won FDA Fast Track designation for pelareorep in later-stage anal cancer, and the drug also holds Fast Track status in colorectal and pancreatic cancer.
From Anal Cancer to an HPV Platform
Chief Executive Officer Jared Kelly framed anal cancer as “the starting point for building a pelareorep plus checkpoint inhibitor platform” across HPV-driven tumors. In addition, he said the survival data and recent regulatory clarity from the FDA give Oncolytics a route to pursue registration while expanding into other indications.
To that end, Oncolytics plans to explore further SCAC studies alongside a potential checkpoint inhibitor partner. The company believes durable responses could support an accelerated approval, while overall survival data could later support full approval, subject to FDA agreement.
Head and neck cancer stands out as the next logical target. HPV drives approximately 70% of oropharyngeal cancers in the United States, and checkpoint inhibitors are already standard therapy there. Pelareorep has shown antitumor signals in earlier head and neck studies, although it has not yet been tested with a checkpoint inhibitor in that disease. As a result, Oncolytics says it may launch such a combination study in the near term.
Why It Matters for Investors
Oncolytics has spent recent months shoring up the commercial foundation around pelareorep. Earlier this month, the company expanded its pelareorep patent portfolio with protection targeting the late 2040s, and in September it reported preclinical support for pairing pelareorep with a pan-RAS inhibitor in colorectal cancer.
Today’s data also lands during a busy stretch for immunotherapy. Earlier today, PRISM MarketView covered how Merck and Moderna’s cancer vaccine results are putting off-the-shelf immunotherapy small caps in focus. Consequently, a partnered registrational path in anal cancer, combined with a potential move into the larger HPV-driven head and neck market, gives Oncolytics several catalysts to watch.
